Research summary
Palmitoyl Pentapeptide-4 (Pal-KTTKS) is a synthetic matrikine: the pentapeptide sequence Lys-Thr-Thr-Lys-Ser is a fragment of type I procollagen, conjugated to palmitic acid to improve lipophilicity and skin penetration. Mechanistically it is reported to act as a feedback signal that prompts dermal fibroblasts to upregulate synthesis of extracellular-matrix components including collagen types I, III and IV, fibronectin, and glycosaminoglycans; this is supported primarily by in vitro fibroblast culture work. The most cited human evidence is Robinson et al. (International Journal of Cosmetic Science, 2005), a 12-week double-blind, placebo-controlled, split-face study in 93 women comparing a moisturizer with 3 ppm pal-KTTKS against the vehicle, which found significant reduction in fine lines/wrinkles by technical image analysis and expert grading, with the peptide being well tolerated. Smaller and manufacturer-associated studies have reported reductions in wrinkle depth, volume, and skin roughness, and a later randomized comparison versus acetyl hexapeptide-3 (Argireline) found both peptides improved crow's-feet over baseline. Limitations are notable: many supporting studies are small, industry-sponsored, or use proprietary blends (e.g., Matrixyl 3000, a distinct combination of palmitoyl tripeptide-1 and palmitoyl tetrapeptide-7), and findings do not always transfer between formulations. There is essentially no rigorous evidence for injectable, oral, or systemic use; the compound is characterized and regulated as a leave-on topical cosmetic ingredient. The U.S. Cosmetic Ingredient Review panel has assessed palmitoyl pentapeptide-4 and related pentapeptides as safe as used in cosmetics, with topical irritation and sensitization reported as low.
